Sample requirements

Quality

Only when samples meet our internal quality criteria, we can guarantee good quality data. These criteria are:

  • Nanodrop OD 260/280 > 1.8

  • Nanodrop OD 260/230 > 1.8
  • Please pay attention that the peak of the Nanodrop profile is situated at 260 nm and not at 270 nm (this would indicate a phenol-like contamination)
  • For high molecular weight (HMW) DNA sample preparation, please consult us as standard wetlab practices may induce DNA shearing or be detrimental to sequencing
  • Note that over 30 samples, you can provide your samples in a PCR plate in column orientation instead of eppendorf tubes.

 

Amount

For DNA applications, 500 ng of genomic DNA will usually be sufficient
Please always provide 3-5 µL extra which is to be used in our quality control steps.

Sample submission

Before sending/bringing your samples, please fill out the appropriate DNA sample submission form and email it to nucleomics.lab@vib.be.
Please use the HMW-DNA sample submission form below for ultra long-read DNA-seq on ONT platform.
Please use one of the PB sample submission form below for long-read DNA-seq on the PacBio platform.

Barcoded primers (PacBio Sequel)

If you wish to multiplex amplicons on the Sequel platform, we prepared a custom barcoded primers Excel file to design up to 16×24 PB-Sequel barcoded primers based on those used in the PacBio 16S application. This primer custom design allows pooling up to 192 pairs (2x 96 well plates) as  found in the worksheet 'bc_primers 192'  while design for up to 384 pairs (4x 96 well plates) can be found in worksheet 'bc_primers 384' of the same file. Please contact us to design such experiment.

When producing your own barcoded amplicons for high-plex sequencing, please use and share with us the Excel files PacBio_Sequel-IIe_amplicon_barcoding_v1.xlsx (up to 2x 96 multiplexed samples per run) or PacBio_Sequel-IIe_amplicon_barcoding_v2.xlsx (up to 4x 96 multiplexed samples per run). Please note that the barcode combinations used in the two files are overlapping but not identical. Please consider using barcode pairs figured in the same worksheet when providing each amplicon plate.
